| CD40L

Cluster Of Differentiation 4 Ligand (CD4L)is a protein that is primarily expressed on activated T cells and is a member of the TNF superfamily of molecules. It binds to CD4 on antigen-presenting cells (APC), which leads to many effects depending on the target cell type. On TFH cells, CD4L promotes B cell maturation and function by engaging CD4 on the B cell surface and therefore facilitating cell-cell communication. A defect in this gene results in an inability to undergo immunoglobulin class switching and is associated with hyper IgM syndrome. Absence of CD154 also stops the formation of germinal centers and therefore prohibiting antibody affinity maturation, an important process in the adaptive immune system.

| Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, wash technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data are provided below for reference only.

Concentration (ng/mL) 10 5 2.5 1.25 0.625 0.312 0.15 0
OD 2.3 1.65 1.06 0.56 0.29 0.17 0.11 0.05
Corrected OD 2.25 1.6 1.01 0.51 0.24 0.12 0.06 -

| Repeatability

Intra-assay Precision (Precision within an assay):CV% < 10%; Inter-assay Precision (Precision between assays):CV% < 10%

| Recovery

The recovery of Human CD40L spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Sample Type Range(%) Average Recovery(%)
Serum(n=8) 84-101 96
Plasma(n=8) 92-105 102
Cell Culture Supernates(n=8) 96-108 105

| Linearity

Samples were spiked with high concentrations of Human CD40L and diluted with Universal Diluent to produce samples with values within the range of the assay.

    Serum(n=4) Plasma(n=4) Cell Culture Supernates(n=4)
1 : 2 Range (%) 85-95 88-96 90-110
Average (%) 91 93 96
